NAD+ (nicotinamide adenine dinucleotide) has become one of the most talked-about molecules in longevity science, showing up in supplement aisles as NMN and NR pills promising to “reverse aging.” The actual research is more nuanced than the marketing — NAD+ decline with age is real and well-documented, but whether boosting it back up meaningfully slows aging in humans is still an open question.

What NAD+ Actually Does

NAD+ is a coenzyme present in every cell, essential for converting food into cellular energy and for activating sirtuins, a family of proteins linked to DNA repair and cellular stress resistance. A review of NAD+ metabolism in aging and disease confirms NAD+ levels decline substantially with age across human and animal tissue — by some estimates, older adults may have roughly half the NAD+ levels of their younger selves. This decline is associated with reduced mitochondrial function, one of several hallmarks of biological aging.

Do NMN and NR Supplements Actually Raise NAD+?

Yes, this part is fairly well established: both NMN (nicotinamide mononucleotide) and NR (nicotinamide riboside) are NAD+ precursors, and human trials have confirmed that supplementing with either reliably raises blood NAD+ levels. Where the evidence gets thinner is the next step — whether raising NAD+ this way translates into the same longevity and metabolic benefits seen in animal studies. Most human trials so far have been small, short-term, and focused on safety and biomarkers rather than actual healthspan or lifespan outcomes.

What the Evidence Actually Supports Right Now

  • NAD+ decline with age is well-documented. This part of the science is solid and not seriously disputed.
  • NMN/NR supplements do raise NAD+ levels in the blood. Confirmed in multiple human trials, generally well-tolerated at studied doses.
  • Direct evidence for anti-aging or longevity benefits in humans is still limited. Most of the striking findings (extended lifespan, reversed muscle decline) come from mouse studies — promising, but not proof the same happens in people.
