L-Theanine and Caffeine: The Science Behind the Focus Stack
L-theanine plus caffeine is one of the few nootropic combos with real human trial data. Here's what the studies on attention, alpha brainwaves, and jitters actually found.

L-theanine and caffeine together form one of the most-studied “stacks” in the nootropic world, and unlike most trendy combinations, this one actually has human trial data behind it. In controlled studies, adding roughly 100-200 mg of L-theanine to a modest dose of caffeine improved attention-switching speed and accuracy, sharpened focus in sleep-deprived participants, and produced a calmer, less jittery experience than caffeine alone. The effect isn’t dramatic, and the research base is still small, but it’s real enough to look at on its own terms, separate from general “brain booster” claims. What Each Ingredient Does on Its Own
Caffeine works mainly by blocking adenosine receptors in the brain, the same receptors that build up “sleep pressure” over the day. Less adenosine signaling means more alertness, faster reaction time, and a subjective feeling of energy. The trade-off is well known: at higher doses, or in people who are sensitive to it, caffeine can also bring jitteriness, a racing feeling, or anxiety.
L-theanine is an amino acid found naturally in tea leaves, particularly green tea. On its own, it doesn’t reliably improve performance on cognitive tests in most trials, but it is consistently associated with increased alpha-band brainwave activity, a pattern linked to a relaxed-but-alert mental state. That’s the property researchers have tried to pair with caffeine’s stimulant effect.
The Proposed Mechanism: Alpha Waves and a Smoother Edge
Alpha-band brain activity
A 2008 study by Kelly and colleagues, published in The Journal of Nutrition, used EEG to measure oscillatory alpha-band activity while participants completed a visual attention task after taking L-theanine, caffeine, the combination, or a placebo (Kelly et al., 2008). L-theanine alone changed brainwave patterns but produced no measurable behavioral improvement by itself. The combined treatment, however, showed lower overall tonic alpha power alongside a real behavioral gain: higher hit rates and better target discriminability than placebo.
Taking the edge off caffeine’s jitters
The working theory is that L-theanine’s calming properties offset some of caffeine’s overstimulation without blunting the alertness gain underneath it. That’s a plausible mechanism supported by the brainwave data above, but it’s still a proposed explanation, not a settled fact. No trial has directly proven that theanine “cancels out” jitters in a measurable, dose-specific way; the calmer subjective experience is inferred from mood ratings and task performance, not a single clean biomarker.
What Human Trials on the Combination Actually Found
Faster, more accurate attention-switching
In a 2008 crossover trial of 27 healthy adults, Owen and colleagues compared 50 mg of caffeine alone, that same dose combined with 100 mg of L-theanine, and a placebo (Owen et al., 2008). Caffeine alone improved subjective alertness and accuracy on an attention-switching task at 90 minutes. The combination went further: it improved both speed and accuracy on the attention-switching task at 60 minutes, and reduced how much participants got thrown off by distracting information during a memory task at both 60 and 90 minutes.
A bigger boost when you’re running on no sleep
A more recent double-blind, placebo-controlled crossover study looked at whether the combination helps when attention is already compromised (Nawarathna et al., 2025, British Journal of Nutrition). Thirty-seven healthy young adults were kept awake overnight, then given 200 mg of L-theanine with 160 mg of caffeine or a placebo. The active combination improved hit rates and target-distractor discrimination on a selective-attention task, and reaction time improved by about 52 milliseconds versus roughly 14 milliseconds for placebo. EEG readings also showed faster neural processing during the task, consistent with the behavioral gains rather than a coincidence.
Taken together, these trials point the same direction: the combination tends to outperform caffeine alone on attention-heavy tasks, particularly attention-switching and sustained focus under distraction or fatigue, more than on raw reaction speed or memory recall in general.
What Ratio Do the Studies Actually Use?
There’s no single official “focus stack” ratio, but the trials cluster around a few patterns worth knowing before you compare labels on a supplement bottle:
- Owen (2008) and Kelly (2008): 100 mg L-theanine with 50 mg caffeine, a 2:1 theanine-to-caffeine ratio, using a caffeine dose roughly equal to a weak cup of coffee.
- Nawarathna (2025): 200 mg L-theanine with 160 mg caffeine, closer to a 1.25:1 ratio, using a caffeine dose closer to a strong cup of coffee, tested specifically under sleep deprivation.
- Common commercial “focus” supplements often market a 2:1 theanine-to-caffeine ratio, echoing the older, lower-dose studies rather than the newer high-dose trial.
In short, the ratio that gets repeated online, roughly twice as much theanine as caffeine, does show up in the original research, but it isn’t the only combination that’s been tested, and the “right” ratio likely depends on your baseline caffeine tolerance and how alert you already are.
The Honest Caveats
This is a more evidence-backed combination than most nootropic trends, but it’s worth being clear-eyed about the limits of what’s actually been shown:
- Small sample sizes. The trials above ranged from 27 to 37 participants. A 2025 systematic review and meta-analysis of L-theanine trials found the overall evidence certainty was “low” to “very low,” largely because of small sample sizes and inconsistent test methods across studies (Mátyus et al., 2025).
- Some industry involvement. Research on L-theanine and tea-derived compounds has, at times, been funded by tea and supplement manufacturers with a commercial interest in positive results. That doesn’t automatically invalidate a study, but it’s a reason to weigh independently replicated findings more heavily than any single trial.
- Modest effect sizes. Reaction-time gains in these trials are measured in tens of milliseconds and accuracy gains in single-digit percentages, real, but not a transformation from “foggy” to “sharp.”
- Mostly single-dose, short-term data. Most trials test one dose on one day. There’s little direct evidence on what happens with daily, long-term use of this exact combination.
- Caffeine sensitivity varies enormously. Genetics (how fast your body metabolizes caffeine), habitual intake, anxiety history, and cardiac conditions all change how a given dose feels. A ratio that feels smooth to one person can feel overstimulating to another, regardless of what a study average shows.
How People Use This Combination in Practice
Green tea and matcha naturally contain both caffeine and L-theanine, though usually in far smaller amounts than the doses tested in trials. A cup of green tea might supply roughly 20-45 mg of caffeine and a modest amount of theanine; matcha runs higher on both. That’s a reasonable everyday starting point if you want a gentler version of the effect.
If you’re trying to approximate what the research actually tested, that generally means a measured supplement rather than tea alone, since brewed tea rarely reaches trial-level theanine doses. As with any caffeine-containing product, timing matters: caffeine’s half-life is several hours, so taking it too late in the day can still affect sleep that night, which in turn undermines next-day focus anyway.
A sensible approach is to start with the lower end of the studied doses, notice how your own body responds, and adjust from there rather than assuming a study average applies to you personally.
