If you’ve read anything about magnesium and brain health lately, you’ve probably noticed two names getting used almost interchangeably: magnesium glycinate and magnesium L-threonate (often sold under the brand name Magtein). They aren’t the same thing, and they aren’t marketed for the same reason. Glycinate is the form people take to relax and sleep. L-threonate is marketed specifically for memory and focus, on the claim that it crosses the blood-brain barrier better than other magnesium forms. That claim traces back to one real, fairly elegant piece of neuroscience, and a much thinner pile of human evidence than most product pages let on.
Where the Blood-Brain-Barrier Claim Actually Comes From
Magnesium L-threonate wasn’t discovered by accident. It was engineered by a team including MIT-affiliated researchers who were specifically trying to solve a real problem: most oral magnesium is absorbed into the bloodstream reasonably well, but very little of it ends up elevating magnesium levels inside brain tissue and cerebrospinal fluid. In the original 2010 study published in Neuron, researchers tested several magnesium compounds in rats and found that L-threonate raised brain and cerebrospinal fluid magnesium more effectively than the others at comparable doses. That difference in delivery, not magnesium itself, is the actual innovation behind the ingredient.
The Foundational Study Was in Rats, Not People
This is the part supplement marketing tends to skip: the striking results people cite (improved learning, working memory, and short- and long-term memory) came from rats, including aged rats that also showed better pattern-completion ability. The same study found higher density of synaptic markers in the hippocampus, the brain region most tied to memory formation, in the treated animals. That’s a genuinely interesting mechanism. It is not the same thing as a controlled trial showing a healthy adult human remembers more names or focuses longer at work.
What Human Trials Have Actually Found
Human data does exist, it’s just smaller and more recent than the animal work. A 2016 randomized, placebo-controlled trial gave the branded compound to 44 adults aged 50 to 70 for 12 weeks and found a significant improvement in a composite cognitive score compared to placebo, though the researchers noted the sample was small and couldn’t draw firm conclusions about sleep or anxiety due to strong placebo effects there. A more recent 2025 randomized trial in 100 adults aged 18 to 45 found modest improvements in overall cognitive performance and reaction time after six weeks, but no measurable benefit on a fluid-reasoning test. Both trials are real and worth taking seriously. Neither is the kind of large, replicated evidence base that lets anyone call the cognitive benefit proven in humans.
How This Differs From the Magnesium You Might Already Be Taking
